Make Crosses & Harvest F1 Seed

This tutorial describes recording and importing crosses into a nursery, advancing crosses to create an F1 harvest list, and recording the F1 seed inventory.

 

Record or Design new Crosses

Export Template

  • From the Crossing Nursery study, select 'Export Crossing Template' from the 'Crossing options' of the Actions menu.

  • Select the disk location to download the Crossing Template.

  • Open the exported Crossing Template from your computer. The crossing template has four worksheets.  The observation sheet contains 2 columns, FEMALE_PLOT and MALE_PLOT, that require data entry for importing crosses. Other details are optional. For more information see Manual: Crossing Template.

Record Crosses

  • We would be making 25 crosses with the crossing nursery list prepared earlier.  The fourth worksheet of the template (Study list) has the details of the entries and the plot numbers in column B.

  • Enter 1 through 25 in the FEMALE PLOT column. Enter 26 through 50 in the MALE PLOT column. Save the changes.  In this example, single crosses will be made between Entry 1 as female and Entry 26 as male, etc.

Import Crosses

  • Select Import Crosses from the Crossing Options tool in the Actions Menu.

  • Browse for and upload the crossing record saved on the crossing template (.xls). Select Continue.

  • Specify Breeding Method. Select Use the parental status to assume the default single cross-breeding method.

  • Continue.

  • Select Use automatic name generation and specify a harvest location. We have used Default Breeding Location as Harvest Location, but you can use Manage Locations Icon to designate location/s specific to your program.

  • Continue.

  • Review pending crosses and select Finish.

Generate F1 Germplasm List

  • Name and describe the F1 Germplasm list. Save.

Saving the F1 Germplasm list opens a new tab in the Crossing Nursery named Imported Crosses.

Harvest F1 Seed

The F1 germplasm list represents the expected offspring from this nursery, independent of actual harvest details. Unsuccessful crosses will result in F1 germplasm with zero harvest amount.

  • Tick on the Select all pages option box and select Create Lots from the Actions menu to record the amount of seed harvested.

  • On the create lots form, enter a stock ID prefix that indicates the 'owner' of the seeds, the 'project' to which they belong, or even 'the storage box' where they will be stored. I specify my initials 'WHE', which indicates 'ownership', and also which section of the seed store they will be kept in.

  • Specify the storage location (which can be a detailed, customized list of shelves or boxes or just a room), indicate the units for recording the inventory, and then specify a dummy deposit of 0.1g so we can have a pending transaction awaiting the actual value when the seeds are weighed.

  • Click Save.

Make labels for the seed packets from a harvest list

The user must use the Manage Inventory item from the INVENTORY menu. Note the user must have a Crop role to be able to access the Inventory Module, otherwise it will not be visible.

  • In the Search by box on the View Lots tab, select 'Study of origin' and click the + symbol.

  • Then click on the new filter item and choose the study where the harvests were made “Crossing Nursery WHE” in my case.

  • This filters the lots down to the 25 F1 seed entries for the harvest list we are working with.

  • From Actions, select Export data and labels. From Output format select Formatted PDF label sheets. Other options are Excel Data or CSV Data which can be used with a printing program such as Microsoft Word – Mail Merge to print labels with any desired format.

  • Select the following fields for the label by dragging the items from the list on the left to the position in the label.

  • Also, choose to have an auto-generated barcode for each lot.

  • You can save the settings for future harvest labels, and you can print the labels for the present batch by clicking Export:

  • Exit the label printing form by clicking Cancel.

Update pending transactions with weights and package and label seeds

  • On the Transactions tab of the Inventory Manager, filter the transactions for the Study of origin– Crossing Nursery WHE (for me) so that I can see the 25 pending transactions.

  • From Actions click Export transactions to get a file of transactions to be updated.

  • Fill in the NEW BALANCE column with the actual weights and specify a new note to replace the original note:

  • Now, we need to import the transaction updates, but this action is unavailable unless the user has a CropAdmin role. If I log out and log in with a crop manager account and then go to Manage Inventory on the transactions tab I can see more options and select Import Transaction Updates:

  • Navigate to the completed transactions update template and import the transactions:

Review Germplasm Details

500 grams of seed has now been applied as available seed to all 25 F1 germplasm entries.

  • Select IB26 to review germplasm details.

  • In the Germplasm Details form, go to the Pedigree tab and notice that the correct breeding method, Single cross, and both female and male GIDs were linked to create this F1 germplasm.

  • Click on the View Pedigree Graph to view the pedigree graph.

  • The pedigree graph shows that the F1 entry IB26 has been derived from a cross between BMS1 & BMS26.
